Our Single Domain VHH library was developed in partnership with Isogenica. After determining that individual camelids only produce 1-10 million unique VHH domains in their repertoire, it was decided that only a synthetic library design could produce the diversity required to take advantage of their CIS-display technology, with library sizes greater than 10 trillion molecules. Through analysis of the fitness landscapes of the camelid repertoire, we developed a synthetic single domain antibody library with display function equivalent to natural camelids, but a diversity 1 million times greater than that found in a camel, llama or alpaca. The resulting library is able to generate low-nanomolar affinity hits with now optimization against any antigen: a perfect library for reagent development and functional characterization against difficult to hit epitopes.
